Of all time? No. In my opinion it's Mario. In the GTA series, I have to go with Tommy Vercetti.

The way he falls down, and gets back up again. Flawless. And the way he acts during cut scenes is beautiful too. How he stares at the camera for long periods of time as he writes what he is saying. Amazing. I give extra kudos to his voice artist. Such skill there.

Claude is the same way. Except he doesn't communicate at all. He just gets the job done.

Yeah, Mike's a champ, I hope we see him again in GTA5 as the main character, only with him still as a 2D top down sprite and everything else in 3D. He'd be the best main character ever for a 3rd person game.

He ain't got much background, unlike almost all others, which makes him somehow mysterious, just like Claude.

So yes, Mike is an excellent protagonist for the GTA series, but whether he's the best - only time will tell.
