Getting in a massive shootout/law enforcement ambush while with a few allies/friends of the protag. deep inside of an airport after arriving in a town would be nice. Starting out with a pistol from a security guard (killed in a melee since you were surprised and unarmed getting off the plane) with a few rounds and picking up weapons from kills along the way. Fighting your way out through swarms of police, TSA, SWAT, and FBI would be difficult given the security levels of American airports these days. It would make for a good mission in GTAV.

A mission involving a firefight in the mountains and forest would be nice too.

I loved the over the top missions in GTA SA. I'd love to see some jets hi jacking or an epic jet chase. Epic chases are my faves.

A car chase mission, where when you almost have the guy and you are coming near a corner a cutscene takes place, you fly around the corner and hit something the car flops sideways hits a telegraph pole and is left smoking, a new camera shot circles around the smoking car and you see your character pulling out his dead friend from the passenger seat.

Always wanted to see some chases where the bad guy gets away.

Someone wants you to kill somebody so you chase them till they hide with a bunch of their boys. Eventually you kill him and drop off your partner.

I have no good ideas for mission plots, but what I would not want to see is repeated missions like in IV, that you had to chase someone, kill enemy gangs, destroy a car, a chopper, a boat...
Make the same thing for different people was annoying.

What I want to see is great finals for missions. Like epic ends for really important missions, with a special kill with something cinematic.
It was added in IV for some missions, but it wasn't that epic. If you ever finished a Modern Warfare game you know what I'm talking. That kind of end, specially in MW2, would be more than amazing for V.

OT:
Missions like Three Leaf Clover would be one of my ideal missions. Pulling any kind of a heist is always the highlight of any GTA game for me, but TLC was especially fun on account of how great the shoot out with the cops was as well as my escape from them through the subway tunnels. It was just awesome.

But it's not always about the action for me, right behind heist missions would be the stealth missions. I just love sneaking around and completing my objectives while trying not to be seen by my enemies. These types of missions can be even more thrilling at times than big shoot outs. Stealth owns.

I'd like a mission where the city ( Los Santos ) is being over run by ZOMBIIESSS!!! and the MC is forced to travel across the map, (on foot) to the other end of the world only to discover he's been bitten half way there. Then the MC wakes up from the dream and the game starts. GREAT PROLOGUE, EH? I think that would be a fantastic way to implement zombies into the game without making it unrealistic!

Don't think it would be nice, dude. It's Grand Theft Auto, not The Walking Dead.
Mixing a GTA campaign with zombies wouldn't be cool in my opinion.

I'd love to rob a bank like in GTA IV then have a huge chase with the cops, FIB, and Military chasing you to the airport, once at the airport you have to steal a plane and try to escape the cops only to be shot down in the countryside by a police chopper and eventually have a huge shootout to end the mission.

I was disappointed by the lame rooftop chases in IV. I'd like to see more of that.

Kidnapping mission or hostage situation mission. Something 50x better than kidnapping Gracie Ancelloti in IV.

Interrogating people is always reall fun for me, like the mission in TBOGT where you beat the crap out of that paparazzi guy then toss him out of the chopper. Not sure if any of you ever played The Punisher game for PS2, but it had brutal interrogation mini games that usually ended in a cool death animation. Not to sound morbid but those were really fun haha so yeah if they included something like that, I'd enjoy it.

Three leaf clover in a bigger scale is basically that mission in GTA SA where you rob the damn caligula's casino. I don't remember it's name but it took like 4 missions just to plan it.

I'd like to see a mission where an entire skyscraper goes up in flames or is leveled in a huge explosion. Very unlikely but basically anything that involves a building getting obliterated will suit me well.
